Buvo parodyta, kad fotobalinimo efektą organinių molekulių dažuose galima sumažinti ir kontroliuoti, kai dažų molekulių eksitonai yra stipriojoje sąveikoje su plazmoniniu sužadinimu. Organinių molekulių fotobalinimo reiškinys yra sąveika tarp sužadintos fluoroforo molekulės tripleto būsenos ir aplinkoje esančio tripleto deguonies. Kadangi tripletinės būsenos gyvavimo laikas yra gana ilgas, sužadintos molekulės turi didesnę tikimybę sąveikauti su aplinkos deguonies molekulėmis ir dėl to įvyksta fotobalinimas. Sąveika tarp molekulinio deguonies ir tripletinių eksitonų vyksta per krūvio pernašos mechanizmus ir sukuria nestabilias reaktyvias deguonies rūšis, kurios gali pakenkti fluoroforams, kas mažina fluorescencijos intensyvumą. Dažų organinės molekulės plačiai naudojamos žymėtų antikūnų panaudojimui imunojutiklių bei greitųjų testų technologijose. Iki šiol nėra atlikta tyrimų analizuojant stipriąją sąveiką tarp žymėtų antikūnų eksitoninių linijų ir plazmoninių sužadinimų. Todėl stipriosios sąveikos tarp žymėtų antikūnų fluorescuojančių dažų eksitonų ir plazmoninių-nanofotoninių struktūrų tyrimai atveria naujas galimybes fotocheminų sąveikų valdyme, kas leidžia pagerinti imunojutiklių detekcijos metodikas. Kai tokia sistema sąveikauja su plazmonine-nanofotonine struktūra, gali būti stipriai paveikti fotobalinimo ir fotoluminescencijos relaksacijos kanalai. Hibridinės plazmon-eksitoninės poliaritoninės būsenos esant stipriajai sąveikai užkerta kelią fotobalinimo procesui žymėtame antikūne. Stiprus elektromagnetinis laukas ir didelis lokalus optinių būsenų tankis, kurį sukelia hibridiniai plazmoniniai rezonansai, gali paveikti cheminius ir fizinius procesus nanometrinėje skalėje. Šio disertacinio darbo metu visiško vidaus atspindžio elipsometrija bus naudojama stipriosios sąveikos tyrimams tarp plazmoninių ir eksitoninių sužadinimų baltymų tarpusavio sąveikų metu.
Theme description
It was shown that photobleaching effect in organic molecule dyes can be reduced and controlled if excitons of dye molecules are in strong coupling regime with plasmonic excitation. Photobleaching of organic molecules effect is caused by interaction between excited fluorophore molecule triplet energy state and triplet of surrounding oxygen. Since lifetime of triplet energy level is long, the excited molecule has higher interaction with surrounding oxygen probability, thus photobleaching occur. The energy exchange between molecular oxygen and triplet excitons occur via charge transfer mechanism and unstable reactive oxygen is created, that can lead to damage of fluorophores. The use of strong coupling between antibodies labelled with fluorescent dyes and plasmonic structures opens up new possibilities for enhancement of rapid test detection limit based on indirect antibody measurement. When such system interacts with plasmonic-nanophotonic structure, photobleaching and photoluminescence relaxation channels can be controlled. Hybrid plasmon-exciton polaritonic states can block photobleaching process in labeled antibody when strong coupling is achieved. The main signature of strong coupling is the energy exchange between plasmons and excitons (in labeled antibodies) that occur during coherence time. Strong coupling regime can be achieved when interaction is strong and energy spectra is modified compared to separate uncoupled excitations of the resonant system. Strong electromagnetic field and high local optical state density caused by hybrid plasmonic resonances can affect chemical and physical processes that manifests in nanometer scale. To the best our knowledge the studies with labelled antibodies in strong coupling regime have not yet been performed. In this Ph.D. the total internal reflection ellipsometry will be used to achieve and study the strong coupling regime between the plasmons and dye excitons in the labelled antibodies.
